Your question is too general and too big in scope AnnieO. Do your research, find what interests you (of which you tell us nothing) and othewise, just play it by ear. A road trip is about what you find along the road and stop for. It is NOT a TOUR where you go from A to B to C etc. following an itinerary. Most people don't really understand what a real 'road trip' is all about.
There is no 'best' route. There is only the route your interests take you on.
Well, Annie0 , I am feeling sad to say that I can't help you in this regard because I have never taken such kind of tour in Europe. I have only been to Paris a year ago with my fiance to enjoy a week tour but your mentioned places are untouched for me.
Bear in mind that Croatia isn't in the Schengen free travel area even though it's in the EU, and going from there to Greece through the Balkans you're likely to go through some non EU countries. Border crossings with your own car may take a long time, and be sure to have your car log book with you as proof of ownership.

London to Edinburgh is perfect place for road trip. It may take around 10 days.
Yes, you could do London to Edinburgh in ten days if you stop at pretty much every place to explore on the way. Alternatively, if there are no jams or road works, you can do it in little over seven hours.
I haven't done a road trip there. But this is what we did last year in case it interests you. Flew in to Brussels, one night in Brugge, drove through Champagne region to Burgundy, stayed two nights in a castle along the Route des Grand Cru. Drove to Switzerland and stayed two nights in Gimmewald. Drove to Alsace and stayed for two nights along the route du vins. Drove to Munich and stayed for two nights during Oktoberfest. Drove back to Brussels and took our flight home. I recommend adding one night in each location as it was a bit hectic. Spend one day driving and two days sight seeing.
