What time of the year? This matters a lot with where to go; the weather can get pretty extreme at various places during various times of the year. Also some general idea of what you're most interested in (cultural activities, adventure activities, hiking, nature, touristy sights, off-the-beat path, wildlife, historical places, etc) helps with giving advice which'll actually be useful for you.

Depending on the answers you give to Sander's questions, I would say firstly it's a huge country so you might have to concentrate only on one part of the country (or be prepared to spend a bit of money to fly). I vote for the Daintree/Cape Tribulation area for natural beauty, Cairns area for the Great Barrier Reef, Fraser Island, the Kimberley's for absolute stunning wilderness, Uluru for true culture and Tasmania. Leave the cities alone- although they are each individually lovely, cities are the same all around the world.
I agree with the answer given by Catsontheprowl.
You will need a few trips to Australia to cover the West and the East sides and Summer is not a great time to travel.
