What are the best things to do in Manizales & Cartagena? Best places to stay? Best tours? Must do restaurants?
Cartagena is by far the most heavily touristed destination in Colombia so it's easy to research.
As for Manizales... it's a pretty cool, laid back town with a bit of a vibe because of all the students there. It would be a nice change after the tourist crunch in Cartagena. I would consider heading south into the coffee region around Salento, there's some beautiful country down there too.

PS Which guide book are you using?
I would also recommend spending some time in the coffee region and staying on a coffee farm if you can - I stayed at one called Hacienda Venecia, not far from Manizales, and it was amazing. The scenery is stunning around there. Salento is definitely worth a stop for a couple of days and from there you can visit the Valle De Cocora which is lovely.
In Cartagena it's nice just to wander the streets and explore - try and get away from the main touristy spots so you can see the little streets without the crowds.
As Terry said, it's a fabulous country. I loved Colombia!
