Are you planning to go by local transport or by "tourist bus"? I am not sure if there are tourist buses that go between the two. If not, there should be tourist buses that go from Bhairahawa or Bhutwal near Lumbini to Pokhara. Tourist buses will be a little more fast. You may want to break up the trip into two days for more comfort if taking local buses. Both ways will be a good full days travel. I am guessing up to 9 to 12 hours depending on road conditions and traffic.

Assuming you are travelling by bus:
Last May, i took a local bus on the opposite direction Pokhara - Bhairahawa (which is about 20min from Lumbini, with frequent bus connections to the site) and the journey took almost 8h. We left Pokhara after 11h and arrived in Bhairahawa around 19h.
Most certainly there is a northbound bus doing the route
