As far as I know and can remember (it's been a while), there are direct buses to Punta Arenas that basically take a whole day (10-12 hours). From there, frequent buses go to Puerto Natales, but I think you have to spend the night again in Punta Arenas, as the last bus might have left then. I believe it's about 3 hours or so to Puerto Natales, so nicer to travel in the morning and enjoy the views anyway.
