Buses can be difficult between countries and take a very long time. Flying though often expensive can be worth it (cheaper if booked as early as possible.)
If you type: bolivia tourism
into a search engine, it will give you the places worth visiting there. You can also check tours, and then visit the places by yourself.
While in Santiago take an hour bus ride to Valparaiso. Peru has most of the places in SA worth visiting, including Lima.
You don't say much about the kind of things you're interested in seeing/doing so it's a bit hard to recommend a route.
Buses in Argentina and Chile are superb - some of the best you're likely to encounter anywhere in the world. So even if the distances are great and the journey times long, they're by no means an endurance test. Peru has some decent buses too, Bolivia not so much ...
Southern Argentina and Chile will be in the middle of the southern hemisphere winter, which may not appeal depending on what your tastes are. My opinion differs to that of Cyberia in that I found most of Peru's main tourist sites to be unattractively over-commercialised - though Arequipa is well worth a visit, and so is Huaraz (though the latter might be a little out of the way for you).
Hi - in 2008 I spent a lot of time in South America, all travelled by bus so you won't have any problems.
Whilst in BA you may want to consider jumping on the ferry over to Uruguay for a couple of days - Montevideo is a bit like a baby BA.
I did a sort of reverse route from your plans, started in Lima, bus down to Arequipa (Colca Canyon), bus to Cusco, if you get the chance stop off in Huacachina,
Puno into La Paz - once my lungs couldn't take anymore I got the bus down to Uyuni and did a fantastic tour of the salt flats. Headed over the border into Chile and San Pedro de Atacama and then headed south. Crossed the border from Chile into Argentina Osorno to Bariloche and then up to BA.
Although I didn't visit Salta or Cordoba in Argentina they come highly recommended. Also, if it's your sort of thing I would recommend Iguazu falls.
Obviously it all depends on what you want to experience.
In Peru (I'm in Lima at the moment) Cruz del Sur and Ormeno bus companies are probably the better ones - they both have websites so you could look at some potential routes.
