"Must Sees" are difficult to say when we don't really know what your interests are.
For wildlife, the national parks in Malaysian Borneo are brilliant, especially Bako. And there's excellent snorkelling there and in the Philippines too. If you want to see tigers, Ranthambore in India is excellent. And there are other great parks in other countries too.
For historical sights, the rice terraces in Batad on the Philippines, the Angkor complex near Siem Reap in Cambodia, the Taj Mahal in Agra, India, the Great Wall near Beijing, China, the Terracotta Warriors in Xi'an, China have got to be at the top.
Then you have Ha Long Bay near Hanoi. Vietnam, the Wagah border ceremony near Amritsar, India (& Pakistan), a sightseeing plane over Everest in Nepal and these only just scratch the surface.
For cities, Singapore, Bangkok, Siem Reap, Phnom, Penh, Ho Chi Minh City, Hoi An, Kuala Lumpur, Varanasi, Shanghai, Manila, Mumbai, Kathmandu ... the list goes on and on for the countries you are visiting.
Perhaps you should narrow your question down to must-see places in each individual country first and then build your itinerary from that. There are lots of resources on this sight and others to help you plan a route.
