Depends mostly on the dimensions of the backpack, but I can't really see a 50 liter one having small enough dimensions, so probably not. (I have a 35 liter one which barely scrapes by, as long as I don't fill the hood.)
Max hand luggage dimensions seem to be (but check this with your specific airline; many ones have slightly different rules) 56x46x25 cm. A bag which would be those exact sizes would have a volume of 64 liter. But I don't know many backpacks which would stick with those 25 cm of depth, while most 50 liter ones would probably be longer than those 56 cm, too.
Also check for weight restrictions on hand luggage. Going over that tends to not be a problem when you have a small looking bag, but if you have a large bag, there's a much higher chance that they'll want to check.
I really doubt it if a 50L pack would make it. My 40L does.
I think you're confused about carry-on luggage... even in Emirates Business Class you're only allowed two pieces of carry-on, the biggest of which can be 22 x 15 x 8 inches (55 x 38 x 20cm) which I doubt your 50 litre is - and even if it can slip by on dimensions the maximum weight can't exceed 7 kg.

Terry: how I interpret Maria's question is that she's flying Emirates to Thailand, and on that trip has a regular (hold) luggage allowance of 30kg. But then she has a separate flight from Thailand to Sydney with a cheap airline which only allows her hand luggage for free, so she's wondering if the same backpack which was in the hold for the first leg could pass as hand luggage on this second (unnamed) airline.
Maria: If it indeed won't fit, just take the hit of paying for hold luggage. Generally with low-cost airlines it's cheaper if you pay for it beforehand, rather than at the airport.
