Not an expert (I don't work for the state department), but I would presume that if you have #2 (a flight/bus/train to Canada) already booked, then you should be fine.
However, I would expect that simply having #1 (a Canadian work visa) is probably not sufficient.
The point of the requirement is that the US state department wants to know that you are planning to leave for sure. #2 makes this fairly clear, #1 does not--and having both #1 and #2 combined is probably the best proof of all.
