Cambodia does not have train travel. The bus is your best option between SR & PP IMO. Here is a of the many companies that run daily buses. You do have a boat option, but it is overpriced and not as convenient.
I would buy the for Angkor. Bayon, Ta Prom are 'must sees'. Allow one morning for sunrise over Angkor and one evening for sunset at is special, but requires separate entrance fee and transport. The is out that way as well. That will take one afternoon of your 3 days.
Taking the bus down to PP, you will have that afternoon to find a hotel/GH and decide what sightseeing you want to do. run tours to just about everything you will want to see. When I travelled from Siem Reap to Phnom Penh and then to Sihanoukville I went with Mekong Express bus. Good reliable service.
