It is a long day, but many make the journey from Samui over to Phuket. The travel shops on Samui will book a ferry-bus or ferry-minivan combo for you. I would take the bus. More room and safer. You want as much metal around you as possible on the Thai highways.
I suggest you stop in first. There are so many beach options close to Krabi town that it is worth a few days IMO. Certainly is worth day tripping. Just idyllic. You might hit as well. Then take the ferry over to for 1-2 days. Then ferry to Phuket. After Railay & Phi Phi, I think you will find Phuket crowded and expensive.

If you're an animal lover then definitely Borneo - Sabah is the place to head with some interesting accommodation in the jungle!
Check this out sabahtourism.com and look for the Orang-utan sanctuary plus the Tortoise Island and if you want a tropical paradise remote island then head to Lankayan Island for diving
