The airport is out of the way, and it takes about 1 hour to get anywhere, so with only 6 hours (and you might want to leave a few of those hours to get back on the next flight), there's not much you can do...you can go to Disney World, or just walk around the city to get a feel for it. I also encourage you to go into a random restaurant and try something out!

The airport express train (less than $15US each way) can get you into the center of the city in 24 minutes. The train is literally inside the arrival area, and it comes about every 10 minutes, so at worst, it will take you about 35 minutes from the arrival gate to downtown. Plan on spending 30-60 minutes going through immigration, changing money, buying train tickets etc... before you get to the train.
Returning to the airport, and assuming that you've already checked your bags through, and just need to go through immigration and security, I would plan to arrive back at the airport about an hour before your flight, just in case there are hold-ups at immigration. If you need to check your bags and all that stuff, give yourself 2 hours just in case. Luckily, HK has got to be one of the most efficient airports in the world, and most of the time you just walk right through. The last time I went through, I got from the train to my departure gate in under 15 minutes. Also consider that Hong Kong / Central Station (the main station in the city, where the Airport Express arrives) is huge, and depending on which exit you come in, it could take you 15-20 minutes just to walk to your platform.
All in all though, you should be able to have 4-5 hours of free time in the city. That's time to do a bit of walking around the city and see a few things. Of course it's not enough time to see it all, so you'll have to choose.
One of the main tourist attractions is to head up to Victoria Peak. This is definitely worth doing if you enjoy getting great views of a city. But, keep in mind that if it is a busy time for tourism, taking the peak tram can take hours in line. Save time buy hopping in a taxi which gets to the top in 15 minutes or so. You'll probably take 1.5 hours out of your day to see this, 2.5 if you want to wander around at the top.
Causeway Bay is a fun place to arrive in if you want to see HK at it's commercial best. From there you can walk West toward Wan Chai. Between the two there are a bunch of local wet markets on the side streets that are always interesting. From there you can hop on the MTR to Central, and stroll around, making your way up the mid-levels escalators for a great selection of restaurants. From there you are a short walk back to HK Station to catch the airport train.
If you have time, take the Star Ferry across the harbour to TST and take a stroll on the Avenue of the Stars where you get another great view of the city. From there you can easily hop on the MTR back to Central to catch the airport train.
Another option, if you don't want to walk so much, is to head straight to Causeway Bay, and hop on the tram for the whole ride to Central. It takes about 20 mins or so, and is a great way to see that part of city, especially if you get up top and up front of the tram.
If markets is more your thing, another option all together is to head over the the Kowloon side. The markets around Mong Kok, such as the ladies Market, gold fish market, flower market, bird market etc.. are really fun to walk around. If you are there in the evening, head to the Temple Street Market. This area is a little more awkward to get to from the airport, but from Mong Kong station you can easily hop on the MTR back to Central (12 mins) or up to Tsing Yi (19 mins), or even get in a taxi to Kowloon Station. All of those three pick up the Airport Express.
Try the Airport Express Line, it takes only twenty-four minutes from Hong Kong International Airport to Hong Kong Station, Hong Kong Island, you can make a short trip, don't miss the gourmet in Hong Kong, especially the dessert.
Hong Kong Disneyland Park is a good choice, it is near the airport and there are metros to get there, very convenient!

two cents worth, if you like shopping and cheap (relatively) hunts, check out http://www.citygateoutlets.com.hk/home/Pages/home.aspx. its a few train stops away from the Airport and have lots of international brands over there.
