Hy lablevs!
I'm stayed in Ushuaia in August/2012 and in the winter some places will be closed. (I'm don't know if stay in your plans, Ushuaia is a city far south, in fact, the southermost city in the world, many travellers seek the city due to this and take the stamp in the passaport !...)
The fastest way is by air, but depend of your toughts, some persons want to know the region deeply, passing throught many cities, in that case is recomended to rent a car, or carefully plan of purchasing bus tickets. In case of bad weather, the snow could close some roads.
The use of second skin under regular clothes is mandatory in case of extreme cold... (not difficult to happen in winter!!!! oh..i'm don't know from where you are, perhaps you're talking from a very cold city and i'm speaking about clothes, in that case my bad!!!)
You're planning go to Perito Moreno glaciar? Nearest cities, depending of your interests could be a interesting places to visit (Rio Gallegos and Puerto Madrin ...)
and have a nice trip!
