When you advise how much time you have for all the destinations on your wish list. Whether you have any particular interests or just want a general tour of highlights. Whether you will arrive after a transatlantic (arrive Madrid, depart Barcelona is that correct?) or other very long flight, I will make a stab at it. Do you not wish to spend any time in Barcelona?
Cheap and comfortable are usually mutually exclusive. Cheapest will be bus but will eat up time. Comfortable will be train which, if booking far enough in advance, can be cheap(ish), so when.
