I took a tour from Medan to Danau Toba (Lake Toba) in July 2011. The ride (a private car) took 4.5-5 hours on 1-2 lane roads, though if our driver wasn't more of a fatalist, it would've taken a bit longer. It was a pleasant escape; we took a boat between Tuk-Tuk (the town on Samosir Island in the lake) and Parapat, the latter of which hosting shops and restaurants which you wouldn't find as easily once on Samosir Island. We learned a bit about the various Batak people and their culture, and enjoyed the nice views along the way.
I also went over to Banda Aceh, way in the north-are you planning on visiting there as well?
