There's a ferry a few times a day between Wellington and Picton. Takes a few hours (3-ish iirc; it's been a while), and costs something like NZD $40. You'll get great views of the Queen Charlotte Sound on the way to Picton. (Taking a car across on it is pretty expensive; luckily most rental car companies allow you to drop off your car at the one end and pick up a new car on the other end.)
(And of course there's the rope bridge taxi drivers will tell you about, but that's only accessible at low tide.)
