THrough Brazil - Rio, San Paulo, Iguzu. 7 - 10days, travel between R to SP is around 3 hour bus. SP to Iguzu is 6hr. so i thought its realistic.
Just so you know, Rio to Sao Paulo is about 6/7 hours on a bus, and Sao Paulo to Iguacu is about 14 hours. Then a further 14/17 hour bus ride to Buenos Aires from there.
Also, if you take the bus from Santiago de Chile to the Peruvian border, it will drop you in Arica (the border town) near the bus station. Get a colectivo taxi from Arica to Tacna (the Peruvian border town) to take you over the border. From Tacna you can get a bus to Arequipa which is about 10 hours, from there you can get buses to Lima, Nazca, Cusco etc.
I'd recommend Mendoza for sure. Go in the summer and do the wine bike ride (you get a pushbike and a map and you can go to all the vineyards and try all the wine they make). There's also skiing nearby in the Andes, or paragliding, mountain trekking and much more.

A few places I would not miss in SA.
Cordoba and surrounding area - Argentina's 2nd city. Heavy student population. Great vibe. La Cumbre for outdoor persuits and nearby is the Che Museum (the house he stayed in as a boy).
Bariloche (Argentina) - completely stunning
Pucon (Chile) - see above.
Both brilliant for outdoor activities in every season.
Torres del Paine (Patagonia) - best trekking I have ever done.
Rurrenabaque (Bolivia) - fabulous wildlife and a great (cheap) experience.
Salt Flats (Bolivia) - stunning. Do the 4 day 4WD jeep trip from Tupiza.
La Paz - crazy city, great hostels, cycle worlds most dangerous road.
