Take a wine tour in the Barossa valley, take the tram out to the beach suburb of Glenelg and spend a couple of days on Kangaroo Island. The Botanic gardens are also worth a stroll around. As you have quite a long time you could also spend 2-3 days travelling the Great Ocean Road.
I'd also highly recommend spending a few hours at the excellent which is a little way out of Adelaide.

You've probably already worked out that Adelaide is very much a big country town. The city itself is very pretty full of what aussies like to call *old* buildings ..
(and stop laughing all you Englishmen, Ive been to the Fighting Cock, I know your idea of *old* is several hundred years earlier)
I live south of the city and the southern beaches and coast are worth a drive..go to Victor Harbor
if you can..you'll see some lovely coast and can take a horse drawn tram over to Granite Island. (where, if youre lucky, youll see penguins and whales)
Then theres Hahndorf, an old German settlers town (Approx 40 kms from Adelaide) complete with original buildings and gardens. The main street is dedicated to tourism and youll find a wide variety of shops and boutiques with unique local produce and unusual gifts. Its a lovely drive through that area... And I recommend the German Arms Hotel for a really good feed.
Strathalbyn, on the Angus River is also worth a visit, a rural setting, easy drive and lots of little antique shops. Again there are many original stone buildings and a beautiful area by the river to rest, enjoy the gardens and relax.

Adelaide is nice cos its much more chilled out than the other main Aussie cities. They have a good market, called the Adelaide Central Market which is worth a wander around. You can get guided tours which include tastings too. You might also like the National Wine Centre. I liked Rundall Mall, its not particularly big, but definitely nice. For a bit of Aboriginal culture see Tandanya. You could go see the pandas at Adelaide Zoo. These are just a few ideas within the city to get you started anyway
I think the replies given above covers almost everything I would have suggested. The Adelaide Zoo is very good these days and the Cleland Wildlife Park is also very good. Manarto Zoo is also worth a look if your not all zoo'ed out. It falls under the open range variety.
Also definitely check out the Adelaide Hills.
The pubs in and around Adelaide are great and the food at the Central Markets is cheap and very good. There are a few nice restaurants around the Mitchem Shops area, along Gouger Street in the City and in North Adelaide.
I also like walking up and down Brown Hill Creek Road - lots of koalas along the way, just look up!
