Since you've never travelled before I would suggest starting with Thailand. It's arguably the easiest "foreign" country for first timers because it has hosted millions and millions and millions of low budget travellers for decades so it has an excellent backpacker infrastructure in place. Start with Khoa San Road and go from there.
You give no clue of your nationality or where you'll be flying from but start Googling embassies of the countries you're interested in so you know Visa entry requirements.

You're going from one side of the world to the other, so you're likely to shell out quite a bit of money for flights, unless you're already in the Americas or in Asia. Suggest you first concentrate on Southeast Asia, then visit South America later, especially if your budget is limited. Also, as CheersT observes, traveling in Southeast Asia is easy. Bangkok is an excellent gateway. If you have six months in the region, you might include Cambodia, Laos and Singapore. Check out the discount airlines, including Air Asia, Cebu Pacific, etc., to see where they fly. You can get information on passenger rail service at seat61.com. Look at a map, then plot a course. If you're determined to go to the Philippines and Indonesia, consider flying from DPS to MNL or vice versa. Several carriers ply that route, so fares are reasonable. If you're going to the Philippines, be aware that you may be required to show that you have transportation out of the country when you go through immigration and customs. Palawan and El Nido are great places to visit.

Your right , I thought SE Asia and South America in 6 months would be too much in one go. My partner would prefer to do South America and I would prefer to do SE Asia. There are so many countries I want to see, I think I am getting a bit over excited to cram in too much in one go.
We have been to Thailand for a two-week holiday - Bangkok, Krabi, Koh Samai however we would like to see more of the country.
Sticking to SE Asia would it be feasible for 6-months without rushing?

If the flights are affordable then SE Asia and South America make for a great combo. The two entirely different cultures are a fantastic travel juxtaposition. It all comes down to budget.

Yes, you could do Southeast Asia leisurely in six months. Plus, it's one of the least expensive regions on the world to visit, with great food, spectacular sights and lots of transportation options. For those reasons, it's very popular with travelers. I recommend setting some priorities for your trip. What do you really want to see, or experience? Then plot your course. Keep the weather in mind, particularly countries that are affected by tropical cyclones, such as the Philippines, Vietnam, Thailand, Cambodia and Laos. The typhoon season runs from May-November. Malaysia, Singapore and Indonesia aren't as affected, since they are near the Equator. You'll find that there aren't inexpensive flights from Southeast Asia to South America. You could potentially get an "affordable" fare from Southeast Asia to Australia (there are lots of Australians flying that route). But the gateway from Australia to South America typically is Santiago, Chile. And the one-way ticket from say, Sydney to Santiago, will cost you a pretty penny. Now, you could double-back and go from Southeast Asia to South America by way of Europe, the Middle East, South Africa or North America, but that will cost you, too. To get an idea, visit some online travel agents. My suggestion remains: Do either Southeast Asia, or South America. But not both, unless you have a generous travel budget.

One more thing. Travel in South America isn't as inexpensive as travel in Southeast Asia. Transportation options are more limited. Check out the airfares between South American countries. You'll find that they are much higher than in Southeast Asia for the distances covered. The same holds true for Africa.
I'll second berner256's assertion that you can spend 6 months in SE Asia and it won't be too much time. I've known travelers who set out to go all over the world, go to SE Asia first, and stay in that region. There is a ton of diversity in the region, and as mentioned above, your money goes rally far there.
If you decide to go to South America, I would recommend spending about 1 month in each country you visit, simply because South America is so big. Giving yourself (on average) a month to see each country would be a good ratio.
Thanks so much for taking the time to respond and help me!! My partner doesn't want to spend 6 months in se asia, and since we're over that way would like to include Australia and new zealand. Therefore what we thought we could do is 1 month Australia, 1 month new Zealand then 4 months in se asia. Does this sound Ok?
Thanks for the advise, your right we need to look at what we want to do in each place then we can work out from there the durations durations. Thanks for giving me ideas of where to start, it's all quite overwhelming and I didn't know where to begin
