Eleven Cities in just 33 days? That's only two to three days per city! Enough maybe for Brighton or Nice but by no means for London, Paris, Rome, Barcelona nor Amsterdam. Even more when you are travelling by train. So I recommend to go by plane e.g. for Amsterdam - Venice. Check out

If you went from London straight to Paris, you could take the eurostar from St Pancras Station and you'd be in Paris within only a few hours. It might also give you more time to explore both cities. And definitely do the Amsterdam-Venice leg by plane if you want to maximise the time you spend in both cities.
The trains can be good if they are direct, they cover a lot of ground very quickly. Great for travel within a country and some countries have great connections between each other. Some do not. They require changes in small towns and all sorts of fooling about and it can take a lot of time and put a bit of pressure on yourself.
There are heaps of budget airlines in UK and Europe, calculate the time, sometimes a plane can take just as long by the time you get to and from the airport. You also have severe luggage restrictions on budget airlines so take that in to account, if you have to drop another 50 euro on luggage it wont be worth it eg. Ryan air is only 15kg. Which even a rucksack weighs more usually.
Try something like skyscanner.net which will give you the prices including the taxes so you get a closer price to reality.
But its def. a good idea to fly between Amsterdam and Venice it might save you quite a bit of mucking around because that is a big journey and through the mountains so it probably goes around and takes a while.
My advice to you would be to compare time and costs. It might be worth saving yourselves the extra time esp. because you have a lot you want to see. You will waste a day on the train unless you do it overnight.
Personally if it came to it, I would leave Amsterdam early to get more time in Venice.
As #2 points out, this is an extraordinary pace if wanting to do real justice to many places. Do you have a checklist and want to tick those boxes? Not interested in finding a vibe anywhere?
All transport has a place and some movements, whatever the transport, will subtract a half to a full day's sightseeing time. Additionally, I suggest you cannot count your arrival or departure days.
Why do you think 'Eurail' is any kind of option? This is an agency which is there to rip you. I hope you have not fallen for the rail pass advertising machine without doing any maths, (but maybe money is no object)?

So I have been trying to plan for a EU trip for two but just need some pointers as to the best mode of transportation between points on our trip. I know Eurail would be the best option but especially from Amsterdam to Venice, would it be better to fly rather than train?
We're leaving Toronto, Canada on Apr 26 and returning from Madrid on May 30. Any help would be much appreciated.

If I were you I wouldn't do Brighton at all. It's beaches have nothing to do with the US ones (never been to Canada but suppose you've ot great beaches too).
I'd go to Florence first after Venice. Or you may consider visiting France first, then Italy and Spain thus they would all be on the way.
Hi phillip, I can give some pointers from Italy.
If you are a wine fan, you should try and get out to the Chianti Classico or Brunello wine regions, worth a splurge on a wine tour if you are short on time as they will book everything for you.
Of course you are probably well versed on the amazing museums in Florence, Rome and Venice - but if you get a chance at least in Tuscany, you should try and get into the countryside.. April is lovely, and there are so many delightful towns outside the main center - Siena and San Gimignano are to be noted... if you need any help, do let me know. happy travels
