One of the hotels I have stayed in, in Siem Reap was the Arboretum. That was OK, within easy walking distance of the main area, and not too expensive.
There is a train from Bangkok (main station on Rama 4, last station on MRT) to Singapore, but it is a long trip with uninteresting views throughout. A quick AirAsia flight is better, and about the same price.

I agree with 'cyberia' on the BKK to SIN leg. Fly it. Many airlines & times to pick from. is usually your best option. Here is a for all things Cambodia. flies from SGN to REP. I would go overland from Siem Reap to BKK. flies this route, but it is expensive. Read about the and then chat with other visitors once in SR. Decide once there. Now that the road is paved, it is not so bad.
