I think probably Sydney and Cairns if you have no restrictions on the other two cities you can visit. From Sydney you can check out all the normal stuff in the city like the harbour bridge and opera house and you can do a day trip into the blue mountains.
From Cairns you could do a reef trip, and perhaps a couple of days up to Cape Tribulation.

The Melbourne Observation Deck
St Kilda
Federation Square
Melbourne Central
Phillip Island Penguin Parade
