Can anyone give any advice about travelling Peru to Brazil overland? Idealy from Cuzco to Rio. Can it be done? Any good bus companies?

You mean directly crossing into Brazil from Peru? In that case check the section here on the tp guide that I wrote. Some small and basic border crossings and you need to travel a long route overland via Porto Velho and further east/southeast.
Otherwise, a better and much more diverse option would be travelling down to Bolivia and to before taking the Death Train to the border with Brazil. From there it's a short hop to the Pantanal and further east to Rio. Have fun.
