It depends on how long you are there for, your budget and how far you want to go up the east coast really.
Hi, I would recommend a trip to Melbourne but that's mainly because I love Melbourne! If you have the time/money you should definitely not miss it out!
I have just travelled the East Coast down from Cairns and we used the Greyhound bus, had no problems to speak of although sometimes you are limited as some places only have a couple of buses a day going through at odd hours that may not be the most convenient. I have also just travelled up from Sydney to Brisbane by train which was ok, although doesn't stop at many of the popular backpacker places and doesn't seem to be a popular way to travel for backpackers probably partly for that reason, it's not ideal for that style for travelling.
A lot of people hire vehicles although we decided against this as we didn't want to have to drive so I can't really offer any advice on that, I guess there are lots of people on here who could though!
