Hello! My girlfriends and I are planning a trip to Europe hopefully in November, and would like to go to London, Amsterdam, Paris, and Berlin. We were thinking three days per city (plus the added travel days to/from the states), but we have no idea which route would be the most efficient. We are open to either flying or taking a train between cities.

Are you flying to London from your home?
If so best route would be fly from London to Berlin, then train to Amsterdam, then train to Paris, then train/flight back to London.
100% the best way of doing it - as coldwarpsy has suggested. Any other way would be almost going backwards and would work out more expensive.
Plus doing it in that order, gives you more flight and train choices and better price options.
You could do.. London - Paris - Amsterdam - Berlin and back to London, but the flight options are not so good - nor are the prices. There are more budget airline flights from London - Berlin, than there are from London - Paris.
It makes perfect sense to do it in the first order suggested.
There's nothing too ambitious about your plans at all. its a very easy trip and wont take long to do either.
The trains from Berlin - Paris, with Amsterdam in the middle are also very good and there are many ways of doing it.
As I've already said, the budget flight options from London - Paris aren't as good as Berlin - London. So that also applies Paris - London. You may want to have a look at what Eurostar has on offer for the dates you need to come back to London. It may very well work out cheaper to take the train, than a one way flight.
