There are quite a few options by ferry to get between HK and Macau. There are even ferries from HK airport to Macau. Check our and/or articles for more info, also about sights and activities.
Regarding time: I suggest a daytrip from HK, while spending all nights in Hong Kong. Also gives a little bit more rest instead of dragging your luggage for a day to Macau.
For Hong Kong: 3 days, (much) more if you are visiting many museums, want to visit some outlying islands or go hiking and exploring the hills and mountains....yes, Hong Kong is much more than just a city.

When you are buying the ticket for the ferry: there are some agent hanging around the official booth offering tickets on a significant discount. These are linked up with the official vendors and they basically collect free tickets from the macau casinos that they can convert into real tickets. Locals often use their services given the cheaper price.
Also in macau, you can use free casino buses/vans to get around.
Macau has some interesting areas that are seldom visited by tourists, they look more like a mix of portugese and mainland chinese towns.
But Mike is right - one day should be enough for macau.

People who say Macau is worth a day, may be because they didn't find cheap accomodations???? People go to Macau, just to hit the airports, I met a few.
I have been to Macau, Three times... Well whats great about it?
Three parts:
1. The Peninsula, with its border with China, has that China town feel, which will not be so appealing to tourist from the 3rd world, traveling on a package tour. This area has lots of churches, if you move further away from the border: Most recommended church is the one on top of the Hill, its called PENA, Lady of Pena... and that other church also on a hill, with a watch tower (very tiny, but its on a hill). Then you have the iconic landmark, that everyone visits, go around 8:00 pm and you will be alone, in what is the most touristic site of the city.
2. The you have the Wynn and Lisboa casino in the edge of the city... that connects to TAIPA island by great bridges... In case you want something 1st world, after wandering in the China town/History tour.
3. Taipa is about Narrow streets, cobbled stones and Candy colored Portugese two storey houses lining up narrow streets... I love it, coz I am Asian, you probably have that everywhere in Europe. So if you get bored, you can go to VEGAS. it's just in the corner... Casinos galore...
4. COLOANE, this is where you can get your cheap accomodation, Bus 25 to Pria Hac Saa, you MUST have the YHA card, coz this hostels is not interested in having guest, they are RICH, government owned, ideally, you booked in advance also, because on weekends they place get booked by the locals. 2 hostels exist in the area, both on the beach...
what people don't know about Macau: It has fantastic TREKKING trails, both in Taipa and Coloane... Walk along the rocky Coastline or the pine wood forrest with two Bluish green lake or dam.... the beaches are most often empty, epic shoreline with a 1st world ambiance.
and I almost forget the area in Coloane that has the St. Xavier church, its also very charming.... Its land of Casinos but it is very QUIET, its like Vegas meets Laos.
well Macau, should atleast be 3 days... HK 2 days... (disneyland not included.)
I just spent 2 weeks in HK, the last time I went there... But I had other things to do...
Macau and HK are so different from each other, its amazing, how its so different.
I like narrow streets and Hills, lots of hills on urban areas, like in Tokyo. Small houses, trendy stores, and the skyline that renders HK a very 1st world ambiance... My favorite part, is the hills and the beaches, where condominiums and international schools are located.
OH....HOng kong is one of most beautiful city in asian whatever the night view, sea view also are amazing. If you ask anyone who tavel in hong kong, they will say awesome, wonderful.....etc. Also, the travellers love shopping in hong kong. And i recommend causeway bay, tsui sha tsui, mong kok.......ohhhh, make girls crazy~~~
p.s i dont hv any idea about Macau....
