Quickest way? Bus, no doubt unless you can afford to pay a private cab (which won't save much time at all, the bus drivers driving the way they do). As I recall, it's only a 3-hour journey or so.
Just a suggestion but I prefer Mindo to Banos and it's only about 2 hours by bus. I've been 4 times to Ecuador totaling over a year and Mindo has much better hiking, waterfalls (that you can jump from), ziplining, etc. Again, bus is best.
