If you drive from Melbourne to Sydney, it will cost you plenty in one way charges, as in someone has to be paid to take the car back where you got it from.
You can hire a car in Sydney for a day trip to Canberra or take a tour.
On the way north I would stop off at the Gold Coast and Brisbane. That is about 500 miles which would give you a break.
Cairns is getting on a thousand miles further. You could of course get a flight from Sydney to Cairns, best booked before arrival.
Greyhound is OK but can quickly become very boring on long journeys.
