No doubt you can find fuel costs via the internet, if not budget for the cost of fuel in the Uk (as it is so expensive here), £1.05/litre, always better to err on the side of the higher cost.
As for car spares, no doubt a motoring association such as the AA will have sensible guides on this subject.

In Turkey gasoline is very expensive it is about 3.2 try per liter that makes around 1.6 euros.
if yo use tem ( e-6 motorway) it is also pay -road but really clean and less crowded than free highways..
There is a bunch of paperwork involved around taking a UK-registered car into Iran and Pakistan and maybe Turkey too. I think you need a carnet de passage. Talk to the embassies.
Visa to Iran are difficult right now if you have UK citizenship. So visa and papers for the car for getting into Iran is the first thing I would sort out. If you are Pakistani the visa for Iran won't be a problem, but then you have to deal with visa in the European countries.
I don't know much about cars, but I think you should be able to get high-quality engine oil and tyres between the UK and Hungary and in Istanbul. After Ankara supply for luxury cars is not so good, but there are plenty of mechanics to help you fix a potential problem.
Tip: Iranian petrol is not so good, consider installing a special filter to deal with that. Iranian petrol is also often sold in Eastern Turkey (because it is only one-tenth of Turkish petrol), so planning to drive all the way from Turkey to Pakistan without a gas stop is not a good solution either.
This sounds like a very intersting trip, but I wonder which way you are driving it.
Austria doesnt border Romania and Romania doesnt border Turkey. But Im sure you know that.
You will have to do lots of planning to get a UK vehicle into Turkey and even more so beyond. Plus getting down to Turkey can be twitchy if you dont take the right route. The shortest route is via Austria, Slovenia, Croatia, Serbia and Bulgaria. However the cheapest route and slightly longer is via Austria, Hungary, Romania and Bulgaria. If you arrive at the Croatia/Serbia frontier without the correct green card insurance then it is expensive for a UK registered car to cross into Serbia. The green card fee into Serbia is €145 minimum. It is very difficult to get Serbia green card cover on a UK motor policy. So avoid Serbia unless you have money to burn.
When you get to the Turkey border (I recommend crossing over from Bulgaria or Greece and not via Ukraine, Russia and Georgia, the paperwork is endless and so painful) there will be another lot of papers to fill in. You need a visa and another green card. It doesnt stop there. The Turkey/Iran border doesnt come in one single drive. First you have to go to the Iran Diplomats Office in Ankara and get another motor document. This can take days and isnt cheap either. When you apply you must have both UK paper and picture driving licences with you along with your already approved and stamped Iran entry visa. If you dont already have a visa then you will not be issued any car documents to cross into Iran, thats it. So you must get a visa from the Iran Consulate of your choice before going to the Diplomats Office to apply for your vehicle papers.
At the Iran/Pakistan border there will be more red tape. You must have your visa to enter Pakistan before you think about crossing into the country. There are roads into Pakistan from Iran marked both Lahore and Karachi. Follow the Karach signs otherwise you will end up at the Iran/Afghanistan border instead of the Iran/Pakistan border. The main crossing point is not far on from Zahedan. From there to the Pakistan border is about 40 miles by road.
Quite interesting that people from Iceland and Zambia do not require a visa to ene=ter pakistan. What a funny ol' world we live in.
If you can convince the border guards that you are only going to be in the Pakistan for a maximum of 24 hours then they might not force you to buy a visa. However you would have probably already got one by the time you get to the border.
You may like to look at www.mdtravelhealth.com as there are a lot of jabs you are recommended to have before you get to Pakistan. They are not compulsory, but strongly advised to thave before you go.
The easy bit is the fuel price list. Dearest in Austria probably and cheapest in Bulgaria but not a lot in it. In the UK its about £1 a litre for 95. In Bulgaria its around 86p and in between the two enroute.
Dont worry about the tyres. There will be so many garages selling part worns on your journey. The oil will be easy to find too. You will need a highviz vest and a first aid kit for many countries.
Anything else you need to ask and I can help then it will be a pleasure to help you.

This is the route all the Turkish living in Germany use when they drive to Turkey for the summer. It is the route that makes the most sense from the UK if you ask me.
Ferry to France, then into Belgium (via Brussels) and down the Rhine Valley on the German side (to avoid tolls in France). Crossing the border to Austria near Salzburg, then Vienna and Budapest. (You'll need a toll sticker for Austria, make sure you buy one at one of the petrol stations between Munich and Salzburg otherwise it will be very expensive.)
One more thing: Bridges across the Danube can be a problem further down. I know that cyclists who cycle to Istanbul from Germany or Austria usually follow the Danube and cross from Romania to Bulgaria at Ruse.
A little tip: You might like the book "The Physician" by Noah Gordon. It is about a English man who travels to Iran in the 10th century AD. The route he takes in the book might inspire you.

I came across your post, albeit an old one. What is your status now? Are you still planning it? Or already travelling?
In case you are still planning it, I have a suggestion. Why stop at Pakistan? It borders India, which is a very colorful country to visit. You can enter via Rajasthan, then travel to any of the major cities from where you can exit to UK. In my opinion, it will be easier for you to work with Indian authorities than in Pakistan. Plus, Indian roads are excellent.

Visa depends on the country’s rules where are you going for.
In my opinion you should take some general parts like engine oil, tyre etc. with you.
