Tickets to South America are hell expensive. You can get a One World RTW fare which will do exactly what you want as American Airlines have a flight from New York - Miami and another from Maimi - Rio De Janeiro (depending on when you are travelling it goes daily, but at less popular times of the year it gets scaled down a little) and if you do want to fly to Chile Lan Chile has one from Rio - Santiago. If you do start in New York then a RTW ticket will require you once in Hong Kong to fly to somewhere like London and then across to New York. Personally if you live in the US I would make an effort to take in some of Europe as well if you aren't on a strict time schedule.

I haven't done any tours overland in South America but have done a bit of overland travel (mostly tourrist places in the regions I have been).

The One World Continent based fare allows you to do as many miles as you want but does limit the One World airlines a bit more. If you go for the mileage based one world fare it may work out cheaper seeing your itinerary doesn't have too many ups and downs all over the placebecause your ticket would otherwise be the 5 continent fare like North America - South America - Australasia/Pacific - Asia - Europe then back to North America which is the 2nd most expensive rtw fare.

As a female travelling on your own I would say that taking a tour through South America was probably a good idea. Loads of companies like Tucan and Oasis Overland cover routes through there so maybe take a look at their websites and get some ideas. You will be with about 20 others so will have loads of company and it's not too school bussey although you will be on a fairly tight schedule.
