There is nothing to do in Colonia but relax. They have a nice old colonial downtown, and a lighthouse you can climb up in, and then you are done. You've got about two hours of sightseeing, at most. I like the town, and once stayed there for over a month, but I was LOOKING to relax and do nothing of any consequence.

You seriously need to think what you want to see or do in just 15 days as getting between all those places you mentioned will take at least 5 days up in travelling on buses.
Although i love South America and even more so Argentina i couldnt just pop over there for 15 days and rush around fitting in as much as possible.
Personally i wouldnt waste my time going to Santiago at all and would only concentrate on Argentina.
Also it depends on whether you want to relax or rush around to see as much as possible.
I'll be honest, aside from some Football(Soccer) matches and a possible Graffiti Tour in Buenos Aires, my itinerary is pretty wide open.
Since someone else brought it up I feel okay about saying this.
I just came from Santiago. Lived and worked there for a year. I consider it wasted time. I really didn´t like the place at all, and I, too, think you´d be better served to skip it. It is a big city with not all that much unique culture, if´n ya ask me and...and you kinda did.

I hate to state that thanks to me having a couple exams that fall during my trip to Argentina, Uruguay, and Chile, my trip's been postponed indefinitely.
However, when one door closes, another one can possibly open. With that said, I'm planning a possible trip to Italy next month for Spring Break!
