10 days is really only long enough for 2 destinations otherwise you are going to spend the whole trip travelling along motorways or in airports.
With a short stay I would suggest 5 nights in Barcelona with day trips to the Costa Brava - and 5 nights in Madrid with excursions to Toledo and El Escorial.
Save Andalucia - Malaga, Seville etc. - for a longer future visit. A trip to Andalucia alone needs at least 14 nights IMO.
Gran Canaria is nothing special so I would forget about that for the time being.

I would like to suggest Barcelona for sure and Picasso Museum, Figueres- Museum Dalí, Basque, San Sebastian, Toledo etc. You can consult the following resources as well for some more destinations:

one more question is that if i wish to travel from barcelona to malaga and then to madrid, then what would be the best way of transport.

10 days is too short. Since you arrive in Barcelona, do not miss the most important sites there: Sagrada Familia, Pueblo Español, Ramblas de las Flores (and the old town), Sardanas dancing, perhaps an excursion to Montserrat. You can fly to Malaga (Vueling) and visit the (horrible) tourist locations there. Personally I prefer flying to Sevilla. Visit Giralda, Alcázar there. In one day you can bus from Sevilla to Córdoba, visit the Mezquita and return to Sevilla. Then fly to Madrid and visit the town and, if you are not yet worn out, Toledo and Segovia.
Malaga is a lovely city and a good base for visiting some of the stunning countryside just a few kilometers inland. The resorts west of the city are built up but Nerja 30 minutes to the east is lovely and retains a lot of Spanish charm. As already said, www.vueling.com offer very well priced flights from Barcelona to Malaga - journey time 65 minutes.
However, I still think 2 destinations is enough for a 10 day stay.
